Summary, etc. | Le concept de réserve cognitive et cérébrale permet d’expliquer l’inadéquation parfois observée entre le niveau d’atteinte cérébrale et les performances cognitives d’individus atteints de la maladie d’Alzheimer. Cette réserve, dont l’établissement reposerait sur l’engagement régulier dans des activités éducatives ou de loisirs, diminuerait le risque de démence de type Alzheimer. Dans le but d’identifier les substrats neurobiologiques qui sous-tendent cette réserve, le modèle murin est largement utilisé. En effet, des souris transgéniques modèles de la maladie d’Alzheimer présentent, après exposition à un milieu enrichi, une progression ralentie de la pathologie, mesurable aux niveaux cognitif et anatomique. Le paradigme d’enrichissement environnemental semble donc adapté à l’étude des processus cérébraux impliqués dans l’expression de la réserve cognitive. Des études récentes ont analysé ces processus aux niveaux cellulaire et moléculaire, et suggèrent notamment l’implication des nouveaux neurones hippocampiques produits dans le cerveau adulte. |

Summary, etc. | Brain reserve and cognitive reserve in Alzheimer's disease: lessons from murine modelsThe concept of cognitive and neural reserve has been used to account for the discrepancy between the degree of brain damage and cognitive performances of Alzheimer's disease (AD) patients. This reserve, built on regular participation in educational or leisure activities, has been linked to a reduced risk of AD-related pathology. In order to identify the neurobiological substrates underlying this reserve, murine models have largely been used. Indeed, AD transgenic mice models exhibit, after environmental enrichment exposure, a delayed progression of the pathology, measurable at both cognitive and anatomical levels. Thus, the environmental enrichment paradigm seems to be appropriate to study neural processes involved in the cognitive reserve. Recent studies have analyzed these processes at the cellular and molecular levels, suggesting in particular, that hippocampal adult neurogenesis could be involved. |
