| Summary, etc. | Les immunothérapies ciblant le B-cell maturation antigen (BCMA) se sont développées de façon rapide et importante dans le myélome multiple (MM) ces dernières années. Elles incluent les cellules T à récepteur antigénique chimérique (CAR-T), les anticorps bispécifiques et les anticorps monoclonaux conjugués (ADC pour antibody-drug conjugate). Dans cet article, nous discutons des principales données cliniques et biologiques existantes sur les ADC. Le premier de classe, le belantamab mafodotin, est un anticorps monoclonal ciblant le BCMA, conjugué au monométhyl auristatine F. L’essai DREAMM2 a permis son approbation par la Food and Drugs Administration et l’Agence européenne du médicament, ce qui en fait la première immunothérapie ciblant le BCMA accessible pour le traitement du MM. Ce médicament est actuellement accessible en France dans un programme de post-autorisation temporaire d’utilisation. Son administration se fait toutes les trois semaines par voie intraveineuse, et nécessite une connaissance précise de ses effets secondaires, notamment ophtalmologiques, afin d’optimiser la prise en charge des patients. |

| Summary, etc. | Immunotherapies targeting B-cell maturation antigen (BCMA) have developed rapidly and significantly in multiple myeloma (MM) in recent years. They include chimeric antigen receptor T cells (CAR-T), bispecific antibodies and monoclonal antibody conjugates (ADC). In this article, we discuss the main clinical and biological data available on ADCs. The first in class, belantamab mafodotin, is a monoclonal antibody targeting BCMA, conjugated to monomethyl auristatin F. The DREAMM2 trial led to its approval by the Food and Drugs Administration and the European Medicines Agency, making it the first BCMA-targeted immunotherapy available for the treatment of MM. This drug is currently available in France under a post-approval temporary use programme. It is administered intravenously every 3 weeks, and its use requires precise knowledge of its side effects, particularly ophthalmological, in order to optimise patient management. |
