Summary, etc. During the differentiation of haploid male germ cells, or spermiogenesis, the organisation of the genome undergoes a profound and unique transformation resulting in extreme genome compaction. This maturation involves a structural transition of the genome from a nucleosome-based structure to a nucleoprotamine-based structure, which is only present in spermatids and spermatozoa. Despite the paramount importance of this fundamental transformation of the genome organisation during the generation of spermatozoa, its molecular basis has long remained completely obscure. Today, thanks to systematic approaches involving the discovery of new histones, their post-translational modifications and the associated factors, as well as large-scale structural and functional studies, we are able to propose the first molecular models explaining the genome-wide transformation of the nucleosome and the establishment of a compact, transportable and functional male genome. This review summarises the major advances made in recent years and our current understanding of the mechanisms driving this unique process in biology.

Summary, etc. Au cours de la différenciation des cellules germinales mâles haploïdes, ou spermiogenèse, l’organisation du génome subit une transformation profonde et unique aboutissant à une compaction extrême du génome. Cette étape implique une transition structurale du génome, depuis une structure dont l’organisation de base est le nucléosome, vers une structure basée sur les nucléoprotamines, uniquement présente dans les spermatides et spermatozoïdes. Malgré l’importance primordiale de cette transformation fondamentale de l’organisation du génome lors de la génération de spermatozoïdes, ses bases moléculaires sont longtemps restées totalement obscures. Aujourd’hui grâce à des approches systématiques impliquant la découverte de nouvelles histones, de leurs modifications posttraductionnelles, et des facteurs associés, ainsi que des études structurales et fonctionnelles de grande envergure, nous sommes en mesure de proposer les premiers modèles moléculaires expliquant la transformation du nucléosome à l’échelle du génome et la mise en place d’un génome mâle compact, transportable et fonctionnel. Cette revue résume les grandes avancées réalisées ces dernières années et notre compréhension actuelle des mécanismes dirigeant ce processus unique en biologie.
