Summary, etc. | The objective of this study was to evaluate the composition of bioactive compounds and the antioxidant activity of virgin olive oil (VOO) from an Algerian variety called «Rougette of Mitidja» from a desert region, as well as its protective effects against titanium dioxide nanoparticles (NPTiO2) induced oxidative stress. The bioactive compounds were analyzed using UV-visible spectrophotometry, while antioxidant activity was tested in vitro using DPPH and in vivo on rats. Twenty-four male Wistar albinos rats were randomly divided into four groups: Control, NPTiO2, NPTiO2 + VOO1 and NPTiO2 + VOO2. The analysis revealed that the VOO contained polyphenols, flavonoids and carotenoids with significant DDPH antioxidant activity at 87.70%. NPTiO2 induced oxidative stress by increasing biochemical parameters and peroxidation of hepatic and renal lipids, as evidenced by a rise in MDA with a decrease in GSH and antioxidant enzyme activities in rats. NPTiO2 also affected the hematological profile with a decrease in red blood cells and hemoglobin. VOO treatment exerted a protective effect by lowering biochemical parameters and increasing antioxidant enzymes compared to the NPTiO2 group. Histological findings confirmed the protective effects of VOO on liver and kidney damages caused by NPTiO2. It can be concluded that monovarietal VOO has a potent protective effect against NPTiO2-induced oxidative stress, which is related to its richness in bioactive compounds. |

Summary, etc. | L’objectif de cette étude était d’évaluer la composition des composés bioactifs et l’activité antioxydante de l’huile d’olive vierge (VOO) d’une variété algérienne appelée «Rougette de Mitidja» provenant d’une région désertique, ainsi que ses effets protecteurs contre le stress oxydatif induit par les nanoparticules de dioxyde de titane (NPTiO2). Les composés bioactifs ont été analysés par spectrophotométrie UV-visible, tandis que l’activité antioxydante a été testée in vitro en utilisant le DPPH et in vivo sur des rats. Vingt-quatre rats mâles Wistar albinos ont été répartis aléatoirement en quatre groupes : Contrôle, NPTiO2, NPTiO2 + VOO1 et NPTiO2 + VOO2. L’analyse a révélé que l’huile d’olive vierge contenait des polyphénols, des flavonoïdes et des caroténoïdes avec une activité antioxydante significative du DPPH à 87,70 %. Les NPTiO2 ont induit un stress oxydatif en augmentant les paramètres biochimiques et la peroxydation des lipides hépatiques et rénaux, comme en témoigne une augmentation du MDA avec une diminution du GSH et des activités des enzymes antioxydantes chez les rats. Les NPTiO2 ont également affecté le profil hématologique avec une diminution des globules rouges et de l’hémoglobine. Le traitement par VOO a exercé un effet protecteur en réduisant les paramètres biochimiques et en augmentant les enzymes antioxydantes par rapport au groupe NPTiO2. Les résultats histologiques ont confirmé les effets protecteurs de VOO sur les lésions du foie et des reins causées par NPTiO2. En conclusion, VOO monovariétal a un puissant effet protecteur contre le stress oxydatif induit par NPTiO2, qui est lié à sa richesse en composés bioactifs. |
