Summary, etc. | Résumé Durant ces dix dernières années, les données structurales et fonctionnelles sur le complexe ARN polymérase du virus respiratoire syncytial, agent majeur des bronchiolites et pneumonies chez les enfants, se sont accumulées. Au-delà de l’intérêt purement scientifique de comprendre comment fonctionne ce complexe polymérase permettant au virus de se répliquer, ces données ouvrent de nouvelles perspectives pour développer des médicaments antiviraux toujours manquants. En effet le complexe polymérase des virus à ARN simple brin de polarité négative n’a pas d’équivalent dans la cellule et représente donc une cible privilégiée pour développer des composés ciblant spécifiquement ces virus. Cette revue fait le point sur l’ensemble des données structurales et fonctionnelles maintenant disponibles pour le complexe polymérase du VRS et les perspectives de développement de composés antiviraux ciblant cette fantastique machine moléculaire. |

Summary, etc. | During the last ten years, structural and functional data on the RNA polymerase complex of the respiratory syncytial virus (RSV), the main cause of bronchiolitis and pneumonia in infants, have accumulated. Beyond the scientific interest of understanding how this polymerase complex works and synthesize viral RNAs, these data open new perspectives to develop antiviral drugs that are still missing. The RNA polymerase complex of single-stranded RNA viruses of negative polarity has no equivalent in the cell and therefore represents a target of choice for developing compounds specifically targeting these viruses. This review presents all the structural and functional data now available for the RSV polymerase complex and the prospects for the development of antiviral compounds targeting this fantastic molecular machine. |
