520			_aLes syndromes lymphoprolifératifs B chroniques présentent une activation constitutive de la signalisation du récepteur B à l’antigène (BCR) qui joue un rôle essentiel dans le processus physiopathologique. Les mécanismes d’activation de la voie sont variables et reposent notamment sur une stimulation antigénique chronique et des mutations au niveau d’intermédiaires et de régulateurs de la voie, responsables d’une activation constitutive d’effecteurs de signalisation et de facteurs de transcription dont le facteur nucléaire κB (NF-κB). Le BCR interagit étroitement avec d’autres voies de signalisation indispensables pour la survie et la prolifération du clone tumoral. La compréhension du rôle de la voie du BCR a permis le développement d’approches thérapeutiques basées sur le ciblage d’intermédiaires de signalisation.
520			_aB-cell chronic lymphoproliferative disorders are a heterogeneous group of mature B neoplasms characterized by constitutive activation of the B cell receptor (BCR) signaling, that is essential in the pathophysiological process. The mechanisms of activation of the pathway are variable and often rely on a chronic antigenic stimulation or on mutations in pathway effectors responsible for the constitutive activation of signaling components and transcription factors including NF-κB. The BCR is at the crossroad with other signaling pathways essential for the survival and proliferation of the tumor clone. Understanding the role of the BCR pathway has enabled the development of therapeutic approaches based on the targeting of signaling intermediates.
